PE-22-28 is a peptide derived from spadin, an endogenous peptide derived from sortilin. It has shown antidepressant-like effects in animal models. Limited human data.

• Intraperitoneal (rodent research) — standard range ~3–4 μg/kg (acute and 4-day sub-chronic mouse studies), Single dose or once daily for ~4 days in published behavioral protocols. Ranges from animal research only; no established human protocol. Parent spadin was often studied near 100 μg/kg in mice; PE 22-28 was more potent on a mass basis. Oral gavage (~1 mg/kg) has also been examined in mice. Human mcg-level claims from non-peer-reviewed sources are not grounded in clinical PK/efficacy trials.

No published human PK. In mice, PE 22-28 and related shortened spadin analogs showed longer behavioral antidepressant duration than parent spadin (activity half-effect reported up to ~14–23 h depending on analog/dose vs ~6–7 h for spadin), despite relatively short in vitro serum stability. Plasma terminal half-life in humans is unknown.
